MOTS-C, short for Mitochondrial Open Reading Frame of the 12S rRNA type-c, is a 16-amino-acid mitochondrial-derived peptide encoded within a short open reading frame of mitochondrial DNA.

Unlike conventional peptides encoded by nuclear genes, MOTS-C originates from the mitochondrial genome, giving it a distinctive role in research examining communication between mitochondria and the wider cell.

Laboratory studies have investigated MOTS-C in relation to cellular energy regulation, mitochondrial stress responses and AMP-activated protein kinase (AMPK) signalling. Research has also examined its influence on folate and purine metabolic pathways, including mechanisms associated with intracellular AICAR accumulation and subsequent AMPK activation.

Under cellular stress conditions, MOTS-C has been investigated for its ability to relocate toward the nucleus and participate in regulation of stress-responsive gene expression. This mitochondrial-to-nuclear signalling behaviour makes it particularly relevant to research involving retrograde mitochondrial communication, metabolic regulation and cellular adaptation.
